How can I tell if I am able to use a hotel room key in Apple Wallet?

Look for a button that says ‘“Add room key to Apple Wallet”’ under your upcoming stays in the app. If the hotel supports room keys in Apple Wallet this will show up as an option automatically when using an iPhone.
